چکیده

The fungus Glomerella cingulata damages a wide range of crops in the tropics and subtropics. This fungus produces an extracellular enzyme, cutinase, which has been implicated in the plant disease process. A cutin monomer, 16hydroxyhexadecanoic acid was used to induce cutinolytic esterase activity during saprophytic growth of G. cingulata. Cutinolytic esterase was induced in cultures grown in cutin monomer but was repressed by glucose. The pattern of cutinolytic activity obtained from the induction and enzyme assays correlates with the results of the Northern blot analyses. The cutinolytic activity and expression from this study demonstrates that the enzyme is tightly regulated by the cutin monomer, 16hydroxyhexadecanoic acid.

Thermal stability engineering of Glomerella cingulata cutinase.

Cutinase has been ascertained as a biocatalyst for biotechnological and industrial bioprocesses. The Glomerella cingulata cutinase was genetically modified to enhance its enzymatic performance to fulfill industrial requirements. Two sites were selected for mutagenesis with the aim of altering the surface electrostatics as well as removing a potentially deamidation-prone asparagine residue. The ...

Heterologous overexpression of Glomerella cingulata FAD-dependent glucose dehydrogenase in Escherichia coli and Pichia pastoris

BACKGROUND FAD dependent glucose dehydrogenase (GDH) currently raises enormous interest in the field of glucose biosensors. Due to its superior properties such as high turnover rate, substrate specificity and oxygen independence, GDH makes its way into glucose biosensing. The recently discovered GDH from the ascomycete Glomerella cingulata is a novel candidate for such an electrochemical applic...

Evaluation of multifarious plant growth promoting traits, antagonistic potential and phylogenetic affiliation of rhizobacteria associated with commercial tea plants grown in Darjeeling, India

Plant growth promoting rhizobacteria (PGPR) are studied in different agricultural crops but the interaction of PGPR of tea crop is not yet studied well. In the present study, the indigenous tea rhizobacteria were isolated from seven tea estates of Darjeeling located in West Bengal, India. A total of 150 rhizobacterial isolates were screened for antagonistic activity against six different fungal...

Associations among parasites of Populus

JONES, B., and NEWCOMBE, G. 1999. Associations among parasites of Populus. Can. J. For. Res. 29: 00-000. Two related F2 families of a Populus trichocarpa x P. deltoides hybrid pedigree were examined in the field for the presence of parasites (three fungal pathogens and five arthropod herbivores), during the third and fourth years of tree growth. The null hypothesis of no associations among para...

Biodiversity and tissue-recurrence of endophytic fungi in Tripterygium wilfordii

A total of 343 endophytic fungal isolates representing 60 taxa including 30 morphotypes were isolated from the different parts of the Chinese medicinal plant, Tripterygium wilfordii. In most cases fungal strains were only identified to genus because species identification was difficult in these speciose genera. Non-sporulating isolates were designated as Morphotypes 1 to 30. The endophytic asse...
